چکیده
The changes in cellular nucleotides accompanying the phosphorylation of 2-deoxyglucose by respiring Krebs ascites tumor cells have been studied. The degradation of ATP by dephosphorylation and deamination to form ADP, AMP, IMP, and inosine in 2-deoxyglucose-treated cells was followed chromatographically and electrophoretically. A rise in cellular AMP concentration, a rise and subsequent fall in cellular IMP, and a steady rise in inosine until it was the predominant 250to 260-m/uabsorb ing compound in the reaction mixture were observed when ascites cells were treated with 2-deoxy glucose. GTP was also broken down in 2-deoxyglucose-treated cells, and one of its breakdown products GDP was identified. A pathway is proposed for the 2-deoxyglucose-induced ATP degradation ha these cells. Hexokinase, adenylate kinase, adenylic deaminase, and 5'-nucleotidase, the

Oxidative Phosphorylation and Ultrastructural Transformation in Mitochondria in the Intact Ascites Tumor Cell
We have examined the ultrastructure of mitochondria as it relates to energy metabolism in the intact cell. Oxidative phosphorylation was induced in ultrastructurally intact Ehrlich ascites tumor cells by rapidly generating intracellular adenosine diphosphate from endogenous adenosine triphosphate by the addition of 2-deoxyglucose. The occurrence of oxidative phosphorylation was ascertained indi...
متن کاملMetabolism of Ascites Tumor Cells. Iii. Effect of 2-deoxyglucose Phosphorylation on Phosphorus Metabolism.
The phosphorylation of 2-deoxyglucose by respiring Krebs-2 ascites tumor cells and the effect of 2-deoxyglucose metabolism on cell orthophosphate, acid-labile phos phate, and adenine nucleotide-phosphate in these cells was investigated. In the presence of 2.0 mM orthophosphate 2-deoxyglucose (2DG) was taken up initially very rapidly, 5.7 /umoles/min/ml of cells, but uptake fell off to a steady ...

certain conditions this delicate balance may be disrupted. For example, during massive ATP dephosphorylation and impaired glycolysis [such as that which follows the addition of 2-deoxyglucose (7), glucose plus iodoacetate (16), or glucose plus oxamate (9)1 , there is an elevation of and then a decrease in AMP and ADP. This is followed by a decrease in AT? and AXP.4 The disruption of the energy ...
متن کاملInduction of Apoptosis by a Combination of 2-Deoxyglucose and Metformin in Esophageal Squamous Cell Carcinoma by Targeting Cancer Cell Metabolism
Background: Both mitochondrial dysfunction and aerobic glycolysis are signs of growing aggressive cancer. If altered metabolism of cancer cell is intended, using the glycolysis inhibitor (2-deoxyglucose (2DG)) would be a viable therapeutic method. The AMP-activated protein kinase (AMPK), as a metabolic sensor, could be activated with metformin and it can also launch a p53-dependent metabolic ch...
متن کاملby D—Glucoseand 2—Deoxy--D--Glucose in Ehrlich Ascites Carcinoma Cells*
Several aspects of the effects of i>.glucose and 2-deoxy-n-glucose on respiratory metabolism of the Ehrlich ascites carcinoma cells are compared. After a brief stimula tory period which was longer for deoxyglucose than glucose, both compounds inhibited the oxygen consumption rate to a similar extent. Glucose-induced inhibition was re leased after all the glucose was utilized, but deoxyglucose-i...
